Why are prodrugs so prevalent in drug discovery?
The #AdvancesChemBio by @gfg-chembio.bsky.social are off to a great start with an insightful workshop by Kurt Ritter addressing this topic.

This 2015 article provided me with food for thought this week "Why is it hard to terminate failing projects in pharmaceutical R&D?" https://www.nature.com/articles/nrd4725 (pay walled, unfortunately). It does a great job summarizing why it is so hard for teams and individuals to stop projects - despite clear opportunity costs and the high failure rate in drug development. The authors suggest 7 concrete steps toward a quick-kill strategy: #Biotech #DrugDevelopment #Science
Loved going back to my science roots and writing about antimicrobials for Drug Discovery News This article summarizes the design of the fully synthetic antibiotic called cresomycin.

Here's an interview with Grigory Khimulya on how we got from vaccine idea to clinic in 6 months at Alvea!
Some highlights:
- Debunking the meme of regulatory red tape that slows innovation
- Lean team instead of siloed departments
- Automation of paperwork where possible
- Look at all vendors in the world, find the best ones and optimize their internal processes as well to fix your bottlenecks

Circular mRNA company Laronde — a real "Moderna 2.0 but also Theranos 2.0" at the same time — merges with another Flagship company amidst data integrity woes:
Until I worked on this piece, I had no idea how out of control blood pressure still is for most people. Estimates are that only one fifth of those with high blood pressure manage it effectively. New RNA drugs that may only require dosing two to four times a year could really help, but at what cost? My latest Clinical Pipeline column.
Interesting, but the article is frustratingly vague about the mechanism of action.
The drug/MoA is probably the same as in this article
"PnPP-19 potentiates erection, in vivo and ex vivo, via NO/cGMP pathway, does not affect sodium channels or rat heart, and shows no toxicity and low immunogenicity, what makes it a very promising candidate as a novel drug in the therapy of erectile dysfunction."

NO is antimicrobial and it's possible this drug could be used for other purposes.
"On Tuesday, an international team announced in the journal Cell that they had identified a potential new antibiotic lurking unnoticed in the sandy soil of North Carolina. The compound, called clovibactin, employs an unusual method of killing bacteria that makes it tough for targets to develop resistance. While clinical trials in humans are several years away, the finding is a glimmer of hope in an increasingly drug-resistant world."

In April I wrote about how this new class of drugs were developed for #NatureMedicine. Read it at the link below:
My new Clinical Pipeline column is out in Nature Medicine.
It's on JAK inhibitors for alopecia - and at the black box safety warning the FDA attached to this entire class of drugs.

Very good news this week with the FDA approval of GSK's RSV vaccine for adults 60 and over - it's the first approved RSV vaccine.

Several other candidate RSV vaccines are at a late stage of development, including mRNA based vaccines. The next big thing to watch for are those seeking to protect newborns and infants by maternal immunisation.
